Compounds this community declines to help with, and why — setting out what I have, and where I think it stops being reliable.

Putting the numbers in the first post, because a question about a compound without them turns into a question about somebody's impression of it.

semaglutide, monoisotopic mass near 4113.6 Da, 25 weeks of my own notes, and 5 lots from 4 suppliers with a purity figure on each. That is the whole basis of what follows.

What I want checked is the reasoning I have built on top of it, not the figures themselves.

f.fenwick, post #5: Thank you — that answers what I came here to find out. Go to post

Post #3 describes the usual case. This is about the unusual one.

Thymosin alpha-1 has a regulatory history in several jurisdictions and a real clinical literature, which puts it in a different evidential category from most of its neighbours here.

It is a small point and it changes the answer, which is an awkward combination.

s.hartmann, post #8: When "no data" is the complete and final answer: if there is no published human data on a compound, that is the state of knowledge. Hope is not a substitute and reasoning from theory is not a substitute. That is not a reason for shame; it is the honest epistemic position. This has been discussed before and I could not find the thread,… Go to post

The tanning compounds carry a specific practical point that is not pharmacological: any change to a pigmented lesion is a reason to see a clinician, and that is not a matter of opinion or of dose.

Research use only, not approved for human use, and in this subcategory the compounds vary enormously in how much is known about them. It is worth establishing which end of that range a specific compound sits at before anything else.

I have deliberately not rounded that, because the rounding is where the argument starts.

b.okonkwo, post #24: Building on post #23 rather than restating it. Anything in this subcategory with a published trial behind it should be discussed separately from anything without one. Mixing them produces a discussion where the confident claims come from the compounds with the least evidence. Marking that as an opinion rather than a finding. Go to post

Dose conversions between related compounds in this subcategory are made far too casually. Structural similarity does not imply potency similarity and frequently does not imply the same receptor profile.

I would rather be precise about what I do not know than vague about what I do.

The arithmetic in post #56 is right; the assumption feeding it is the part to check.

Where a compound has a legitimate pharmaceutical form somewhere in the world, the labelling for that form is usually the single most useful document available and is almost never consulted here.

That is all the detail I have. Someone else will have more.

Second this, and I would have said it less carefully.

Compounds this community declines to help with, and why: there are specifics in the guidelines and they are not arbitrary. They exist because of failure modes that have happened in real people, not because of prudishness.

That is the honest state of it as of this week.
