Does a peptide degrade silently, with no visual change? — that is the question, and I have not found it answered plainly anywhere I have looked.

I have read the maintained page on this and I still have a gap, so I am asking rather than guessing.

Context: tirzepatide, 6 weeks in, currently at a dose I reached by the standard four-week steps. Everything below is my own record rather than anything a clinician told me.

The specific question is the one in the title. What I have already checked: the labelling summary on the relevant documentation page, the two most-linked topics in this subcategory, and my own notes from the last 14 weeks. What I could not find is whether the answer changes at higher doses or whether it is the same arithmetic throughout.

If the answer is "it depends", I would rather know what it depends on than be given a number.

Visual inspection: if a solution is visibly cloudy, discoloured, or contains particles, those are red flags. Faint opalescence or minor colour shift may be normal but if you are unsure, the supplier can answer specifically.

I had written a reply contradicting post #2 and deleted it. Here is what survived.

The failure mode on peptide is boring rather than dramatic. It is almost always the step everyone assumes was done correctly because it is too simple to get wrong.

Practical answer on peptide, since the theoretical one is upthread: do the simplest check first, write down the result, and only then decide whether the complicated explanation is needed. It usually is not.

The version of peptide that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed.

Someone should write this up properly, and it should probably not be me.

Answering the question post #6 raises rather than the one it answers.

Two people in this thread mean different things by peptide and are disagreeing about the definition while believing they are disagreeing about the facts. Worth pausing to define it.

Coming back to post #9, because the follow-up matters more than the original answer.

Adsorption to container surfaces removes peptide from solution without changing how the solution looks. In dilute preparations that loss can be a meaningful proportion, and it is invisible.

I have left out the parts I could not verify.

Post #9 is right about the mechanism and I think understates the practical bit.

What I would check first on peptide is whether the thing being measured moved or whether the way of measuring it moved. Those look identical in a graph.

Peptide was covered in the wiki last year and the page has a review date on it, which is a better starting point than my memory of a thread.

A definition problem is doing most of the work in this peptide discussion. Once the term is pinned down I suspect the disagreement mostly goes away and what is left is small.

Picking up post #17: that is the part I would want checked first.

Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed.

Where I would look next, rather than where I would stop.

The arithmetic on peptide is the easy part and it is where the errors are, which is an uncomfortable combination. Show your working and someone will catch it.

Freeze-thaw: some peptide solutions tolerate freezing poorly because ice crystals damage protein structure. Whether your specific solution tolerates it depends on the formulation and is not something to test empirically. The conservative position is not to freeze.

Someone will know this better than I do and I hope they say so.

Something worth flagging about peptide: the strongest-sounding claims in this thread are the ones with no source attached, which is the usual pattern and not a coincidence.

I read post #28 twice before replying, because I had assumed the opposite.

Container choice matters: the material of the vial, the rubber closure, the fill volume all affect how quickly the contents degrade. Smaller fill volumes lose less to adsorption. Glass vials lose less to leaching than plastics.

It is worth checking rather than assuming, which costs nothing.
