Counter-ion form: the field almost nobody reports — one year on

On the subject in the title: Counter-ion form: the field almost nobody reports — one year on Working notes rather than a conclusion.

The question about Counter-ion form that I actually want answered is the second one below. The first is context and I have kept it short.

Both are stated with units, and I have said what I already checked so that nobody repeats it.

Reading back through the Counter-ion form threads from last year, the same three questions come up every time and only one of them has ever been answered properly. That seems like a documentation gap rather than a knowledge gap.

Post #2 is right about the mechanism and I think understates the practical bit.

Counter-ion form is well covered in the tag pages, and the older discussions are better than the recent ones because they were argued out properly. Worth twenty minutes before adding to this one.

Coming back to the opening post, because the follow-up matters more than the original answer.

An itemised related-substances table with retention times says considerably more about the synthesis than a single total. A total tells you how much is not the main peak and nothing about what it is.

If anyone has run this properly I would rather read that than my own guess.

A stability statement tied to a named storage condition can be evaluated. One without a condition attached cannot be evaluated at all and is decoration.

The distinction between release testing and characterisation is one industrial documents make and retail ones usually do not. It is the difference between "we tested every lot for this" and "we established this once".

That is the shape of it. The detail is where I would expect to be corrected.

Post #6 answers the question as asked. The question underneath it is different.

If someone has run Counter-ion form properly I would rather read that than my own reconstruction of it. Posting mine only because the thread has gone quiet.

Agreed on Counter-ion form, with one qualification that I think matters. The reasoning holds for the case as described. Change the starting assumption and it does not, and the starting assumption is the part nobody states.

The arithmetic in post #6 is right; the assumption feeding it is the part to check.

A chromatogram supplied as a small image is legible for peak shape and not for baseline detail. That is enough to sanity-check an integration and not enough to reproduce it.

If anyone can point at the primary source I would be grateful.

Answering the question post #8 raises rather than the one it answers.

Two people in this thread mean different things by Counter-ion form and are disagreeing about the definition while believing they are disagreeing about the facts. Worth pausing to define it.

Counter-ion form is crucial: peptides are isolated as salts, most often trifluoroacetate from reversed-phase purification or acetate after salt exchange. The counter-ion is part of the mass in the vial and is not part of the peptide. Two vials of the same purity, one as TFA and one as acetate, contain different amounts of peptide per milligram.

Posted with less confidence than the sentence structure implies.

I would call the community position on Counter-ion form likely rather than established, and I would be comfortable defending that hedge.

Adding the measurement that post #11 says would settle it.

Counter-ion form is a question about a distribution, not about a value, and treating it as a value is what produces the confident wrong answers.

Post #15 put the caveat in the right place and I want to underline it.

The honest answer on Counter-ion form is that it depends, and the useful part is the list of what it depends on. Four items, in rough order of how much they matter.

Most people get the first two right and then argue about the fourth.

Post #20 is right about the mechanism and I think understates the practical bit.

I disagree with the framing of Counter-ion form above, and I think it is a substantive disagreement rather than a terminological one. Setting out why, so it can be checked.

The reasoning depends on an assumption that is doing a lot of work and is never stated. If the assumption holds, the conclusion follows. I do not think it holds generally.

Purity and content are different measurements: purity tells you what proportion of the detected material is the intended species. Content tells you how much of the intended species is present in the container. A lyophilised vial can be 99% pure and contain considerably less than the label claims because the remainder is water, counter-ion and excipient.

On reflection I would soften that slightly.

The useful distinction on Counter-ion form is between what was measured and what was inferred from it. Both end up in the same sentence and only one of them has error bars.

The fields that make one usable: lot identifier matching the container, test date, method identification specific enough to reproduce, the result as a number, and the acceptance criterion stated separately from the result.
