Melanocortin agonists: mechanism and the documented adverse profile — setting out what I have, and where I think it stops being reliable.

Melanocortin agonists, and specifically the version of it that the documentation does not cover. The maintained page handles the general case well and stops exactly where my question starts.

Setting out the gap in case it is a gap in the page rather than a gap in what is known.

What I would want before treating melanocortin agonists as settled: the method, the sample, and whether anyone tried to find the opposite result. Two of the three are usually missing.

Solubility varies far more across this subcategory than within the incretins. A compound that needs a specific diluent or a specific pH is not being difficult; it has a solubility profile and the supplier should state it.

If anyone has run this properly I would rather read that than my own guess.

Reading a supplier's product description as a marketing document: that is exactly what it is. The hyperbole, the mechanism written as fact, the theoretical benefits stated as established benefits — all of that is marketing. Treat it as such and separate it from evidence where evidence exists.

Not a conclusion. A place to stand while looking for one.

Answering the melanocortin agonists question as asked, then the question I think is meant. As asked: yes, with the qualification below. As meant: it depends on how the first measurement was taken.

Melanocortin agonists: mechanism involves the melanocortin-4 receptor pathway that regulates appetite. The documented adverse profile includes blood pressure elevation and erections of sustained duration, the second of which is specific enough that it is the first thing worth mentioning.

Written quickly, so the reasoning may be tighter than the wording.

Building on post #6 rather than restating it.

The question underneath melanocortin agonists is usually "how would I tell?" rather than "what is true?", and that one has a method attached to it.

Write down what you would expect to see under each hypothesis before you collect anything. If they predict the same observation, collecting it will not help.

AOD-9604 is a fragment of human growth hormone and has been discussed as a potential weight-loss agent based on theoretical mechanism. The fragment hypothesis — that the fragment retains a specific property of the intact hormone — is not settled for this compound.

The claim is narrower than it sounds, and deliberately so.

Picking up post #8: that is the part I would want checked first.

PT-141 was developed from the same family with a different receptor profile, and it has a considerably better documented human evidence base than most compounds discussed in this subcategory.

If the premise is wrong, everything after it is decoration.

Confirming post #8 from a second method, which matters more than confirming it from a second person.

Two questions I would want answered before drawing anything from the melanocortin agonists data above: how were the cases selected, and what happened to the ones that dropped out.

I had written a reply contradicting post #10 and deleted it. Here is what survived.

Thymosin alpha-1 has a regulatory history in several jurisdictions and a real clinical literature, which puts it in a different evidential category from most of its neighbours here.
