Robustness: the parameters worth deliberately varying Writing it up because I had to work it out twice and would rather nobody else did.

A narrow question about robustness, deliberately narrow, because the broad version has been asked here four times and produced four long threads and no answer.

One question, stated units, stated method, and what I have already ruled out.

A methods point on robustness rather than a substantive one: if the comparison is not like for like, the difference you are measuring is the difference in method.

This follows the opening post rather than contradicting it.

System suitability is the ongoing evidence that a validated method is still performing. Validation is done once; suitability is done every run, and it is the one that appears on a certificate.

That holds for the case as described. Change the assumptions and it may not.

Genuine question rather than a rhetorical one: has anyone here actually observed robustness, as opposed to read about it? The thread is long and I cannot tell.

Post #5 is right about the mechanism and I think understates the practical bit.

Robustness: the method gives consistent results when minor parameters vary. Tested by deliberately varying pH, temperature, flow rate, and mobile phase composition within reasonable ranges and demonstrating that results stay within acceptance.

That holds under the stated conditions and I have stated them.

The arithmetic in post #9 is right; the assumption feeding it is the part to check.

The most useful single question about a method: what would it fail to detect? Every method has an answer and few documents state it.

Building on post #10 rather than restating it.

Robustness is one of those subjects where the general answer and the answer for a specific case diverge, and the thread will go in circles until someone says which one is being asked for.

Post #8 put the caveat in the right place and I want to underline it.

For anyone finding this later: the short answer on robustness is that it depends on one thing, and the rest of the thread is people identifying which thing.

Precision has two levels worth distinguishing: repeatability within a run and intermediate precision across days, analysts and instruments. The second is where most methods lose the numbers people quote.

Two things can be true about robustness at once: the mechanism is plausible and the evidence for the size of the effect is thin. Most of the argument here is people defending the first against attacks on the second.

Before the thread moves on from robustness — what is the sample size behind the claim? I am not being difficult; I have seen the same figure quoted from an n of four and from an n of four hundred.

The distinction between a qualified instrument and a validated method is worth keeping. Both are needed and they fail in different ways.

That is my reading. Someone else read the same page differently and was reasonable.

Answering the question post #16 raises rather than the one it answers.

Where I have landed on robustness, having got it wrong once in public: the direction is clear, the magnitude is not, and anyone quoting a precise magnitude has borrowed it from somewhere that did not measure it.

Where a pharmacopoeial monograph exists, a method that follows it inherits a great deal of assurance. Almost nothing discussed here has one.

This is where my knowledge stops and I would rather mark the edge than blur it.

The limit of quantitation determines what the impurity table can honestly contain. Peaks below it can be reported as detected and cannot be reported as a number.

What I would want before treating robustness as settled: the method, the sample, and whether anyone tried to find the opposite result. Two of the three are usually missing.

Confirming post #21 from a second method, which matters more than confirming it from a second person.

My experience of robustness contradicts the reply above. I am posting it as a data point rather than as a refutation, because one person's experience is exactly that.

A method that reports the same number for every lot is worth a second look. Real processes vary, and a total absence of variation is a statement about the measurement rather than the process.

A weak preference rather than a position.

Taking robustness seriously for a moment rather than deflecting: the honest position is that the community has observations and no controlled comparison, and those two things support very different sentences.

Forced degradation studies: deliberately stress the material with acid, base, oxidant, heat, light to generate degradation products and demonstrate that the method can separate them from the parent peak. Acceptance is that the method is stability-indicating.

I would want the raw data before agreeing with my own summary of it.
