On the subject in the title: Second pass at: Robustness: the parameters worth deliberately varying Working notes rather than a conclusion.

Something about Robustness does not reconcile and I would like a second pair of eyes before I decide which half is wrong.

Two sources, both reputable, giving figures that cannot both be right unless they are measuring different quantities. My suspicion is that they are, and I cannot see how.

Adding a reference point for Robustness. Mine is a single case, collected without controls, and I am posting the method alongside it so it can be discounted appropriately.

The failure mode on Robustness is boring rather than dramatic. It is almost always the step everyone assumes was done correctly because it is too simple to get wrong.

Coming back to the opening post, because the follow-up matters more than the original answer.

Where a pharmacopoeial monograph exists, a method that follows it inherits a great deal of assurance. Almost nothing discussed here has one.

The most useful single question about a method: what would it fail to detect? Every method has an answer and few documents state it.

A weak preference rather than a position.

Robustness testing deliberately varies the parameters most likely to drift — organic percentage, pH, temperature, flow — and shows the result does not. It is the part of validation that predicts whether a method will transfer.

That matches what I was told, which is not the same as knowing it.

Summarising the Robustness thread so far, since it is long and the answer is buried: the first reply has the method, the fourth has the correction to it, and the rest is people agreeing at length.

I read post #4 twice before replying, because I had assumed the opposite.

What I can speak to on Robustness is narrow, so I will keep it narrow rather than generalising from it. Beyond that boundary I do not know.

The limit of quantitation determines what the impurity table can honestly contain. Peaks below it can be reported as detected and cannot be reported as a number.

The variance between people here is larger than the effect being discussed.

The most useful reply I ever got about Robustness was a request to state my units. It sounds like pedantry and it has saved me twice.

The distinction between a qualified instrument and a validated method is worth keeping. Both are needed and they fail in different ways.

Written quickly, so the reasoning may be tighter than the wording.

Answering the question post #11 raises rather than the one it answers.

What I would tell a new member reading about Robustness for the first time: the confident posts are not the reliable ones, and the reliable ones are longer.

The strongest argument against my own position on Robustness, stated as well as I can state it, since nobody else has yet.

System suitability is the ongoing evidence that a validated method is still performing. Validation is done once; suitability is done every run, and it is the one that appears on a certificate.

One of those cases where knowing the mechanism does not help the decision.

Forced degradation studies: deliberately stress the material with acid, base, oxidant, heat, light to generate degradation products and demonstrate that the method can separate them from the parent peak. Acceptance is that the method is stability-indicating.

Noting that the question and the thing people usually mean by it are different.

I think the Robustness question is answerable and has not been answered, which is a more optimistic position than most of this thread.

Post #15 and I disagree about the size of the effect, not about the direction.

Two claims get bundled together under Robustness and they need separating. The descriptive one — this is what was observed — is usually well supported. The causal one — this is why — usually is not.

Almost every disagreement in threads like this one dissolves once you say which of the two you are making.

Worth separating two things that post #18 runs together.

What I would check first on Robustness is whether the thing being measured moved or whether the way of measuring it moved. Those look identical in a graph.

A method that reports the same number for every lot is worth a second look. Real processes vary, and a total absence of variation is a statement about the measurement rather than the process.

I would be cautious about generalising from the Robustness example above. It is a good example. It is one example.

What is documented is narrow. What is inferred from it is broad. The gap between them is where every argument here lives.

An observation about Robustness that I cannot explain and am posting anyway, on the principle that unexplained observations are more useful public than private.

Robustness: the method gives consistent results when minor parameters vary. Tested by deliberately varying pH, temperature, flow rate, and mobile phase composition within reasonable ranges and demonstrating that results stay within acceptance.

That is my reading. Someone else read the same page differently and was reasonable.

The version of Robustness that I was taught turned out to be a teaching simplification. Useful, and not true in the way I had assumed it was.

One caution on Robustness: everything above assumes the underlying documentation is what it claims to be. That assumption is doing real work and is rarely stated.
