On the subject in the title: Vagal afferents and the gut-brain pathway Working notes rather than a conclusion.

Vagal afferents — I have the observation and I do not trust my interpretation of it, so I am posting the observation and holding the interpretation back.

Numbers, method and the conditions under which they were collected are below. Interpret them however they warrant.

Signalling through cyclic AMP is the canonical pathway and is not the only one. Beta-arrestin recruitment differs between ligands and its clinical significance here is unestablished.

What I would check first on vagal afferents is whether the thing being measured moved or whether the way of measuring it moved. Those look identical in a graph.

I read post #4 twice before replying, because I had assumed the opposite.

Filing a mild objection to the consensus on vagal afferents. Mild because I might be wrong; an objection because nobody has addressed the case that does not fit.

Half-life extension by albumin binding through a fatty acid chain trades free fraction for duration. It is an engineering solution with a cost, and the cost is that the bound fraction is not active.

The variance between people here is larger than the effect being discussed.

Species differences in receptor pharmacology are substantial in this family, which is one reason rodent data has translated unevenly.

Written in the hope of being told what I have missed.

Post #6 is the version of this I will quote in future. One addition.

Reading this vagal afferents thread as someone who came in with a fixed view: the third and seventh replies moved me and the confident ones did not.

On vagal afferents, I would rather understate and be corrected upward than overstate and be quoted. That is a house style here and it is a good one.
