Storing lyophilised material long term: what governs the shelf life — setting out what I have, and where I think it stops being reliable.

Two things I would like separated before anyone answers on Storing lyophilised material long term, because they get bundled and then argued about as one thing.

The first is descriptive: what has actually been observed, by whom, and how. The second is causal: why. I am asking about the first only.

Confirming post #2 from a second method, which matters more than confirming it from a second person.

Reconstituted material carried in a bag for a day is a temperature excursion even in a cool climate. Where that matters is cumulative exposure over weeks rather than a single afternoon.

I would want a second opinion before relying on that.

The arithmetic in post #4 is right; the assumption feeding it is the part to check.

Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed.

I am describing what is, rather than arguing for what should be.

Worth separating two things that post #7 runs together.

Dry storage in a freezer is generally more protective than a refrigerator for long periods, provided the material is genuinely dry and the container genuinely sealed. Condensation on removal is the trap.

This has been discussed before and I could not find the thread, so, again.

Temperature excursions: if a vial sat at room temperature for a time, the question is whether the excursion was long enough to cause damage. Hours at room temperature is usually low risk. Days to weeks raises the risk. If you are unsure, contacting the supplier is more informative than guessing.

That is a description of practice, not a recommendation of it.

Container choice matters: the material of the vial, the rubber closure, the fill volume all affect how quickly the contents degrade. Smaller fill volumes lose less to adsorption. Glass vials lose less to leaching than plastics.

It is one reading of the data and not the only reasonable one.

Worth separating two things that post #11 runs together.

The arithmetic on Storing lyophilised material long term is the easy part and it is where the errors are, which is an uncomfortable combination. Show your working and someone will catch it.

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

Not the answer, but possibly the question that gets there.

A request rather than an answer: could whoever has the primary source for Storing lyophilised material long term post it? I have seen the claim three times this month and each version had lost a qualifier.

Speaking only to Storing lyophilised material long term as I have actually seen it, rather than as it is usually described: the effect is real, it is smaller than the thread suggests, and the variance between people is larger than the effect.

Protecting from light: amber vials or opaque containers reduce light exposure. If you are storing for long periods and light matters, this is worth doing. For short-term prepared solutions, ordinary storage away from direct sunlight is usually adequate.

A partial answer, offered because a partial answer beats none.

Building on post #23 rather than restating it.

Lyophilised material is far more stable than the same material in solution. That is why suppliers ship it dry, and it is the reason to reconstitute only what will be used.

I would want to see it done twice before believing it once.

Post #23 answers the question as asked. The question underneath it is different.

The two questions that get merged here are whether material is degraded and whether it is contaminated. Storage affects both and they need different answers and different tests.

Posted with less confidence than the sentence structure implies.

On Storing lyophilised material long term, the part that usually goes wrong is that the question is asked as though it has one answer. It has a range, and the width of the range is the interesting bit.

If you can post the two or three numbers you are working from, several people here will check the arithmetic rather than argue about the conclusion.

Door storage in a refrigerator is the warmest and most variable position in the appliance. A shelf at the back is a materially different environment.
