BPC-157: what the rodent literature actually shows, and what it does not — setting out what I have, and where I think it stops being reliable.

I would like to know what people here actually do about BPC-157, as distinct from what is usually recommended. Those have diverged in every other subject I have looked at closely.

Mine is below, with the reasoning, including the parts I am not confident about.

Confirming the opening post from a second method, which matters more than confirming it from a second person.

On BPC-157, I would rather understate and be corrected upward than overstate and be quoted. That is a house style here and it is a good one.

Post #3 is right about the mechanism and I think understates the practical bit.

BPC-157's published evidence is overwhelmingly preclinical and mostly rodent. That is not a dismissal — it is the state of the literature, and any discussion that starts elsewhere is starting from something that is not there.

I keep a log of this specifically because memory is unreliable about it.

Worth separating two things that post #3 runs together.

One caution on BPC-157: everything above assumes the underlying documentation is what it claims to be. That assumption is doing real work and is rarely stated.

The version of BPC-157 that I was taught turned out to be a teaching simplification. Useful, and not true in the way I had assumed it was.

The lyophilised cake is worth looking at before adding diluent. A collapsed or shrunken cake is not a purity finding but it does say something about how the vial was made and how it travelled.

The thing about BPC-157 that took me longest to accept is that a plausible mechanism is not evidence of an effect. It is a reason to look, not a result.

BPC-157 is a good example of a question where the honest answer is boring and the interesting answers are unsupported. I would go with boring.

Taking post #12 at face value and following it one step further.

Reading a preclinical wound-healing model: the model shows whether a mechanism is plausible in a specific context. It does not show magnitude of effect in humans, does not show safety profile in humans, and does not show whether the effect survives in a more complex biological system. That is not a criticism of preclinical work — it is what preclinical work is for.

Whatever the answer on BPC-157 turns out to be, the method for getting there is the same: state the assumption, do the arithmetic in public, invite the correction.

Everything in post #15 holds. The case it does not cover is the one I have.

I have been on both sides of the BPC-157 argument in this category within eighteen months, which should tell you how strong the evidence for either side is.

Building on post #16 rather than restating it.

BPC-157: the preclinical literature is extensive, mostly from a small number of research groups, and reports effects across a wide range of injury models. The breadth of reported effects is itself worth noting — a compound that improves outcomes in tendon, muscle, gut, nerve and bone through a single mechanism would be remarkable, and remarkable claims deserve proportionate scrutiny.

Noting that the question and the thing people usually mean by it are different.

Dose-response is unstudied in humans for essentially everything in this family, which means the confident numbers in circulation came from somewhere other than a trial.

Written quickly, so the reasoning may be tighter than the wording.

Confirming post #16 from a second method, which matters more than confirming it from a second person.

The preclinical work is genuinely interesting and it is preclinical. A rodent tendon model is a model of a rodent tendon, and the translation record from that kind of model is poor.

The strength of my opinion here exceeds the strength of my evidence.

TB-500 is usually the 7-residue actin-binding fragment of thymosin beta-4, not the full 43-residue protein. The two are routinely conflated in supplier documentation and in the literature. Evidence about the full protein does not automatically apply to the fragment.

The answer changed when I changed how I was measuring, which was informative.

Where I part company with post #19, and it is a narrow parting.

Two claims get bundled together under BPC-157 and they need separating. The descriptive one — this is what was observed — is usually well supported. The causal one — this is why — usually is not.

Almost every disagreement in threads like this one dissolves once you say which of the two you are making.

Post #23 is the version of this I will quote in future. One addition.

On BPC-157, the part that usually goes wrong is that the question is asked as though it has one answer. It has a range, and the width of the range is the interesting bit.

If you can post the two or three numbers you are working from, several people here will check the arithmetic rather than argue about the conclusion.

Stability of BPC-157 in solution: what has been measured in published work covers specific formulations under specific conditions. Extrapolating to a reconstituted preparation in a different diluent at a different concentration is an extrapolation, acknowledged as one.

That holds for the case as described. Change the assumptions and it may not.

Publication patterns in the BPC-157 literature: a large proportion of the evidence comes from one research group. That is not necessarily wrong — one group can do excellent work — but it is worth noting when evaluating the breadth of support for a claim.

I would be interested in a counterexample if anyone has one.

I had written a reply contradicting post #23 and deleted it. Here is what survived.

Speaking only to BPC-157 as I have actually seen it, rather than as it is usually described: the effect is real, it is smaller than the thread suggests, and the variance between people is larger than the effect.

The useful distinction on BPC-157 is between what was measured and what was inferred from it. Both end up in the same sentence and only one of them has error bars.

Picking up post #27: that is the part I would want checked first.

BPC-157 is one of those subjects where the general answer and the answer for a specific case diverge, and the thread will go in circles until someone says which one is being asked for.
