The arithmetic in post #5 is right; the assumption feeding it is the part to check. Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed. I have changed my mind on this once already, so take it as current rather than settled.

Does a peptide degrade silently, with no visual change? I have a specific reason for asking rather than idle curiosity, and the context is below.

What changes if the standard account of peptide is wrong? I ask because I have been treating it as settled and I noticed this week that I could not say why.

Working through the consequences rather than the evidence, since others here are better placed on the evidence.

Answering the question the opening post raises rather than the one it answers.

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

That holds under the stated conditions and I have stated them.

Freezing a reconstituted peptide solution is not automatically protective. Freeze-thaw cycling is one of the harsher things you can do to a protein in solution, and a single freeze is different from repeated ones.

That has been true for the cases I have seen and I have not seen many.

Practical answer on peptide, since the theoretical one is upthread: do the simplest check first, write down the result, and only then decide whether the complicated explanation is needed. It usually is not.

Adding a reference point for peptide. Mine is a single case, collected without controls, and I am posting the method alongside it so it can be discounted appropriately.

Freeze-thaw: some peptide solutions tolerate freezing poorly because ice crystals damage protein structure. Whether your specific solution tolerates it depends on the formulation and is not something to test empirically. The conservative position is not to freeze.

I would be interested in a counterexample if anyone has one.

Peptide is well covered in the tag pages, and the older discussions are better than the recent ones because they were argued out properly. Worth twenty minutes before adding to this one.

Visual inspection: if a solution is visibly cloudy, discoloured, or contains particles, those are red flags. Faint opalescence or minor colour shift may be normal but if you are unsure, the supplier can answer specifically.

Worth reading the earlier posts in this thread before acting on mine.

Taking peptide seriously for a moment rather than deflecting: the honest position is that the community has observations and no controlled comparison, and those two things support very different sentences.

Agreed on peptide, with one qualification that I think matters. The reasoning holds for the case as described. Change the starting assumption and it does not, and the starting assumption is the part nobody states.

A methods point on peptide rather than a substantive one: if the comparison is not like for like, the difference you are measuring is the difference in method.

Container choice matters: the material of the vial, the rubber closure, the fill volume all affect how quickly the contents degrade. Smaller fill volumes lose less to adsorption. Glass vials lose less to leaching than plastics.

A modest claim, modestly supported.

Peptide is a question about a distribution, not about a value, and treating it as a value is what produces the confident wrong answers.

I had written a reply contradicting post #25 and deleted it. Here is what survived.

The version of peptide that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Confirming post #29 from a second method, which matters more than confirming it from a second person.

Adsorption to container surfaces removes peptide from solution without changing how the solution looks. In dilute preparations that loss can be a meaningful proportion, and it is invisible.
