Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

I would put this at better than even and not much better.

Practical experience of peptide, offered as one case with the conditions stated, not as a general finding. Conditions first, because they are what make it interpretable.

NLoughran, post #5: Freezing a reconstituted peptide solution is not automatically protective. Freeze-thaw cycling is one of the harsher things you can do to a protein in solution, and a single freeze is different from repeated ones. That has been true for the cases I have seen and I have not seen many. Go to post

Picking up post #32: that is the part I would want checked first.

Container choice matters: the material of the vial, the rubber closure, the fill volume all affect how quickly the contents degrade. Smaller fill volumes lose less to adsorption. Glass vials lose less to leaching than plastics.

On post #30 — agreed on the reasoning, with one qualification.

The reason peptide is hard to answer is that the obvious measurement and the relevant quantity are not the same thing, and substituting one for the other is silent.

Adding what did not work for me on peptide, since the failures never get written up and they are half the useful information.

A qualification I should have led with rather than closed on.

Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed.

Anyone with a larger sample, please post it.

Visual inspection: if a solution is visibly cloudy, discoloured, or contains particles, those are red flags. Faint opalescence or minor colour shift may be normal but if you are unsure, the supplier can answer specifically.

One case, stated as one case.

Post #47 is the version of this I will quote in future. One addition.

One more thing on peptide that took me far too long to see: the two figures people quote are not measuring the same quantity. Once you notice that, the apparent contradiction disappears.

What I can speak to on peptide is narrow, so I will keep it narrow rather than generalising from it. Beyond that boundary I do not know.

I would be cautious about generalising from the peptide example above. It is a good example. It is one example.

Worth separating two things that post #52 runs together.

A definition problem is doing most of the work in this peptide discussion. Once the term is pinned down I suspect the disagreement mostly goes away and what is left is small.

Peptide: I would want to see the raw numbers rather than the summary before agreeing. Summaries lose exactly the information that would settle this.
