Reconciling a label claim with a content assay Writing it up because I had to work it out twice and would rather nobody else did.

A follow-up question about Reconciling a label claim that I did not know to ask the first time.

The earlier thread answered what I asked. What I should have asked is below, and I think it is the one that matters.

Adding the measurement that the opening post says would settle it.

What I would check first on Reconciling a label claim is whether the thing being measured moved or whether the way of measuring it moved. Those look identical in a graph.

The distinction between release testing and characterisation is one industrial documents make and retail ones usually do not. It is the difference between "we tested every lot for this" and "we established this once".

That has held every time I have looked, which is not the same as always.

An itemised related-substances table with retention times says considerably more about the synthesis than a single total. A total tells you how much is not the main peak and nothing about what it is.

I looked this up rather than remembered it, which is the right order.

The arithmetic in post #5 is right; the assumption feeding it is the part to check.

Reading this Reconciling a label claim thread as someone who came in with a fixed view: the third and seventh replies moved me and the confident ones did not.

The version of Reconciling a label claim that I was taught turned out to be a teaching simplification. Useful, and not true in the way I had assumed it was.

A chromatogram supplied as a small image is legible for peak shape and not for baseline detail. That is enough to sanity-check an integration and not enough to reproduce it.

That is what I would do. It may not be what is correct.

Water content (residual moisture) matters: lyophilised peptide is not dry. A few percent water is normal. The difference between a TFA salt and an acetate salt can be several percent. Content calculations need the water content to be accurate.

It is the sort of thing that seems obvious in retrospect and was not at the time.

My understanding of Reconciling a label claim is a few years old and may have been superseded. If it has been, I would genuinely like to know rather than keep repeating it.

Speaking only to Reconciling a label claim as I have actually seen it, rather than as it is usually described: the effect is real, it is smaller than the thread suggests, and the variance between people is larger than the effect.

I had written a reply contradicting post #12 and deleted it. Here is what survived.

I disagree with the framing of Reconciling a label claim above, and I think it is a substantive disagreement rather than a terminological one. Setting out why, so it can be checked.

The reasoning depends on an assumption that is doing a lot of work and is never stated. If the assumption holds, the conclusion follows. I do not think it holds generally.

Water content by Karl Fischer is worth having on a lyophilised product because residual water is both a mass contributor and a stability factor.

Reading it again, the caveat matters more than the finding.

Post #14 is right about the mechanism and I think understates the practical bit.

For anyone finding this later: the short answer on Reconciling a label claim is that it depends on one thing, and the rest of the thread is people identifying which thing.

I keep a log for Reconciling a label claim specifically because my memory of it turned out to be systematically wrong in one direction. Six weeks of notes cost nothing and settled it.

A note on scope: what I am saying about Reconciling a label claim applies to the case in the first post and I would not extend it further without checking.

Post #21 is the version of this I will quote in future. One addition.

Amino acid analysis gives content: it hydrolyses the peptide and quantifies the residues. It measures the peptide content independent of chromatographic purity. Very few supplier certificates report it.

That is a description of practice, not a recommendation of it.

The most useful thing anyone has posted about Reconciling a label claim in this category was a table of what had been measured and by whom. That is what I would want again.

Reconciling a label claim has been discussed here with more heat than it deserves, mostly because two definitions have been in play the whole time.

Peptide content and chromatographic purity are separate determinations and a certificate that reports only one has answered only one question. Content is the one that tells you how much is in the vial.

I changed my mind about Reconciling a label claim after someone here asked me for the source and I could not produce one. That is worth saying out loud because it is the ordinary way it happens.

Narrowing post #25, because the general version has more than one answer.

Two claims get bundled together under Reconciling a label claim and they need separating. The descriptive one — this is what was observed — is usually well supported. The causal one — this is why — usually is not.

Almost every disagreement in threads like this one dissolves once you say which of the two you are making.

Coming back to post #25, because the follow-up matters more than the original answer.

It is a small point and it changes the answer, which is an awkward combination.

What would change my mind on Reconciling a label claim is a second dataset collected by someone with no stake in the first. Until then I hold it loosely and I would rather say so than pretend to more.
