Freeze-thaw and why one cycle is not the same as five — a second dataset — setting out what I have, and where I think it stops being reliable.

Posting a small dataset on Freeze-thaw. It is mine, it is uncontrolled, and the method is stated so it can be discounted appropriately.

What I would like is not agreement but a second dataset collected by someone with no stake in mine. If one exists I would rather read it than argue for this one.

The opening post put the caveat in the right place and I want to underline it.

Freeze-thaw is one of those subjects where the general answer and the answer for a specific case diverge, and the thread will go in circles until someone says which one is being asked for.

Practical note on Freeze-thaw: write down what you expect before you look. The number of times I have found what I went looking for is higher than chance would allow.

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

The mechanism is plausible, which is not the same as established.

Before the thread moves on from Freeze-thaw — what is the sample size behind the claim? I am not being difficult; I have seen the same figure quoted from an n of four and from an n of four hundred.

Two things can be true about Freeze-thaw at once: the mechanism is plausible and the evidence for the size of the effect is thin. Most of the argument here is people defending the first against attacks on the second.

Confirming post #8 from a second method, which matters more than confirming it from a second person.

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

It is one reading of the data and not the only reasonable one.

A beyond-use date on a home preparation is an inference rather than a measurement. It can be a reasonable inference and it should be described as one.

Post #13 is right about the mechanism and I think understates the practical bit.

The reason Freeze-thaw is hard to answer is that the obvious measurement and the relevant quantity are not the same thing, and substituting one for the other is silent.

Freeze-thaw: I would want to see the raw numbers rather than the summary before agreeing. Summaries lose exactly the information that would settle this.

The honest summary: keep it dry until you need it, keep it cold once it is not, keep it dark, do not cycle the temperature, and treat any period at room temperature as spent whether or not anything visible happened.

It is the sort of thing that seems obvious in retrospect and was not at the time.

Freezing a reconstituted peptide solution is not automatically protective. Freeze-thaw cycling is one of the harsher things you can do to a protein in solution, and a single freeze is different from repeated ones.

An honest declaration on Freeze-thaw: I have a prior here and it is strong enough that you should weight what I say downward. Stating it rather than hiding it.

Aggregation is the failure mode that most concerns anyone who works with peptides professionally and the one least visible to anyone who does not. It is also concentration-dependent.

The general answer and the answer for your case may diverge here.

That is the version I would defend. It is not the version I started with.

The practical version of Freeze-thaw is three sentences long. The rigorous version is three pages and reaches the same conclusion with the conditions attached.
