Ipamorelin selectivity claims and where they came from — setting out what I have, and where I think it stops being reliable.

Ipamorelin selectivity claims, from the point of view of someone who has read the documentation and is still stuck.

Saying which parts I have understood so that nobody wastes time re-explaining them, and where exactly the understanding stops.

Development history is more informative than speculation: ipamorelin was investigated clinically for post-operative ileus and was not taken forward to registration. That it was not brought to market tells you something about the cost-benefit calculation that any amount of forum enthusiasm does not.

I read post #3 twice before replying, because I had assumed the opposite.

A request rather than an answer: could whoever has the primary source for ipamorelin selectivity claims post it? I have seen the claim three times this month and each version had lost a qualifier.

A mass result for a short peptide is more discriminating than for a long one, because a single residue difference is a larger proportion of the total. That makes identity confirmation genuinely useful here.

The part I am sure of is shorter than the part I have written.

The useful distinction on ipamorelin selectivity claims is between what was measured and what was inferred from it. Both end up in the same sentence and only one of them has error bars.

Ipamorelin selectivity claims is one of those subjects where the general answer and the answer for a specific case diverge, and the thread will go in circles until someone says which one is being asked for.

Ipamorelin is usually described as selective in that it produces less of an effect on cortisol and prolactin than earlier compounds in the family. Selective is a relative claim and the comparison group matters.

Filing this under things that are true until someone shows me otherwise.

Two people in this thread mean different things by ipamorelin selectivity claims and are disagreeing about the definition while believing they are disagreeing about the facts. Worth pausing to define it.

Ipamorelin selectivity claims originate in preclinical characterisation and are reasonably well supported for what they claim: less effect on cortisol and prolactin than earlier GHRPs. Whether that translates to something clinically meaningful is a different question from whether the selectivity is real.

The claim is narrower than it sounds, and deliberately so.

Why this subcategory is stricter about sourcing than most: the evidence base is weaker and that is precisely why documentation quality matters more. A supplier page that clearly identifies the compound and the purity is more valuable for secretagogues than for incretin agonists where the evidence base is stronger.

Adding it in case it saves somebody the afternoon it cost me.

Taking ipamorelin selectivity claims seriously for a moment rather than deflecting: the honest position is that the community has observations and no controlled comparison, and those two things support very different sentences.

Injection-site reactions are reported more often in this family than for the incretins. Whether that is the compounds or the preparations they are typically supplied in is not established.

I had written a reply contradicting post #18 and deleted it. Here is what survived.

On analysis: these are short peptides and generally straightforward chromatographically, which means a poor purity result is more likely to reflect the synthesis than the method.

Second-hand on ipamorelin selectivity claims, so weight it accordingly — someone whose method I trust told me this and I have not verified it myself.

Effects reported at the level of appetite are the most consistently described and the most mechanistically direct for the ghrelin-receptor compounds, which is not what most people are taking them for.

Reconstitution volumes in this family are often small enough that dead volume in the syringe is a meaningful fraction of the dose. A fixed-needle insulin syringe is worth the trouble here specifically.

Something worth flagging about ipamorelin selectivity claims: the strongest-sounding claims in this thread are the ones with no source attached, which is the usual pattern and not a coincidence.
