Journal club: orforglipron phase 2 and the non-peptide question posts 121–133

This is a continuation of a long topic, addressed by post number rather than by page. Start at post 1.

f.haddad, post #118: The most common failure mode is spending fifty minutes on the effect size and ten on the population. Reversing that ratio would improve most sessions. It is one reading of the data and not the only reasonable one. Go to post

The failure mode on orforglipron phase 2 is boring rather than dramatic. It is almost always the step everyone assumes was done correctly because it is too simple to get wrong.

Where the group cannot agree, record the disagreement rather than resolving it by seniority. The recorded disagreement is more honest and more useful later.

Worth saying I have only my own numbers here, and n is small.

Post #122 answers the question as asked. The question underneath it is different.

Critical appraisal template: (1) What did the trial set out to estimate? (2) Could the design answer that question? (3) Was the population sufficiently similar to your population to apply the results? (4) What was the absolute effect, not just the relative one? (5) What are the two strongest criticisms available?

c.castellanos, post #81: The claim about orforglipron phase 2 upthread is stronger than its source supports. I have read the source. The source says "associated with" and the post says "causes". Go to post

I read post #121 twice before replying, because I had assumed the opposite.

Practical answer on orforglipron phase 2, since the theoretical one is upthread: do the simplest check first, write down the result, and only then decide whether the complicated explanation is needed. It usually is not.

Sessions on negative or null results are consistently the most instructive and the hardest to get anyone to attend.

I am reporting what happened, not recommending it.

Bringing the previous paper on the same question makes the session much better and doubles the preparation. Worth doing every third session rather than every one.

This is the sort of exchange that makes the archive worth searching.

p.iyer_pharmd, post #96: Adding a null result on orforglipron phase 2. I looked, carefully, and found nothing, and null results deserve posting precisely because they never are. Go to post

The version of orforglipron phase 2 that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Counterpoint on orforglipron phase 2, offered without confidence: the same observation is consistent with a much duller explanation, and nobody has ruled the dull one out.

Good question, well framed, and I would like to see it answered properly.

Post #128 put the caveat in the right place and I want to underline it.

People arriving without having read it should be welcome and should say so, because a question from someone who has not read it frequently exposes an assumption everyone else made.

That is what the documentation says. What happens in practice is usually close.

A request rather than an answer: could whoever has the primary source for orforglipron phase 2 post it? I have seen the claim three times this month and each version had lost a qualifier.

Source for the orforglipron phase 2 figure, since it was asked for. It is in the discussion rather than the abstract, which is why the version circulating is stronger than the paper is.

Reading the surrounding paragraph is worth the two minutes. The authors are more careful than their summarisers.
