Preservatives: function, testing, and limits — setting out what I have, and where I think it stops being reliable.

An honest uncertainty about preservatives rather than a disguised assertion.

I do not know the answer and I have not been able to find one. What I have is the shape of the question, which may be worth more than my guess at the answer.

Adding the boring version of preservatives, because the interesting version keeps getting posted and the boring one is usually right.

Check the ordinary explanations, in order, and stop when one of them accounts for what you are seeing. Most of the time the second one does.

Light sensitivity varies enormously between peptides depending on the residues present. Tryptophan-containing sequences are the ones where it matters most.

Preservatives: solutions for multi-dose vials contain preservatives (benzyl alcohol, phenol, or others) to inhibit microbial growth after the vial has been entered. Single-use vials do not need preservatives.

For what it is worth, the same held on the two occasions I checked.

Excipients and their purposes: mannitol, sucrose, or other sugars in lyophilised formulations protect the peptide during drying and storage. They are not impurities; they are functional components.

Confirming post #7 from a second method, which matters more than confirming it from a second person.

Practical note on preservatives: write down what you expect before you look. The number of times I have found what I went looking for is higher than chance would allow.

Surfactants such as polysorbate are added to prevent adsorption and aggregation at interfaces. In a bare research preparation there is nothing performing that function.

If the premise is wrong, everything after it is decoration.

Source for the preservatives figure, since it was asked for. It is in the discussion rather than the abstract, which is why the version circulating is stronger than the paper is.

Reading the surrounding paragraph is worth the two minutes. The authors are more careful than their summarisers.

The reasoning is more useful than the number, which is why I have shown it.

The question underneath preservatives is usually "how would I tell?" rather than "what is true?", and that one has a method attached to it.

Write down what you would expect to see under each hypothesis before you collect anything. If they predict the same observation, collecting it will not help.

Post #14 and I disagree about the size of the effect, not about the direction.

Overfill in a vial is deliberate and accounts for the volume unrecoverable at the end. It is not generosity and it is not a bonus dose.

A single observation, in a thread that deserves better than single observations.

Aggregation is concentration-dependent, which is why reconstituting to a high concentration is not always the convenient choice it appears to be.

Marking that as an opinion rather than a finding.

Two things can be true about preservatives at once: the mechanism is plausible and the evidence for the size of the effect is thin. Most of the argument here is people defending the first against attacks on the second.

Sterility and stability are separate properties requiring separate controls. A preservative addresses one of them and nothing about the other.

One more caveat and then I will stop qualifying: the sample selected itself.

Everything in post #19 holds. The case it does not cover is the one I have.

I read the earlier replies on preservatives twice before writing this, because I had assumed the opposite and wanted to be sure I was disagreeing with what was said rather than what I expected.

The honest answer on preservatives is that it depends, and the useful part is the list of what it depends on. Four items, in rough order of how much they matter.

Most people get the first two right and then argue about the fourth.

Worth separating two things that post #23 runs together.

Counter-ion exchange from trifluoroacetate to acetate is a purification step with a cost, and material sold without it retains TFA. That affects mass balance and can affect biological assays.

Stating my assumptions rather than smuggling them in.

On preservatives I would separate what is worth knowing from what is worth acting on. The first list is long and the second is short, and conflating them is how threads get heated.
