Revisiting: A field-by-field walk through a certificate of analysis Writing it up because I had to work it out twice and would rather nobody else did.

I would like to disagree carefully with the settled view on field-by-field walk through a certificate, and I would like to be argued out of it if the disagreement is bad.

The disagreement is about one step, not about the conclusion. If the step holds I withdraw it entirely.

The opening post answers the question as asked. The question underneath it is different.

Field-by-field walk through a certificate came up in a thread eighteen months ago and was answered well. I cannot find it, which is itself the problem, so here is the reconstruction.

A chromatogram supplied as a small image is legible for peak shape and not for baseline detail. That is enough to sanity-check an integration and not enough to reproduce it.

Everything in the opening post holds. The case it does not cover is the one I have.

The distinction between release testing and characterisation is one industrial documents make and retail ones usually do not. It is the difference between "we tested every lot for this" and "we established this once".

That is where I would start, not where I would stop.

I read the earlier replies on field-by-field walk through a certificate twice before writing this, because I had assumed the opposite and wanted to be sure I was disagreeing with what was said rather than what I expected.

Taking post #5 at face value and following it one step further.

A certificate is a statement by its issuer that a defined test was performed on a defined lot and produced a defined result. That is all it is, and it is not nothing.

Happy to be the one who is wrong here if it settles the question.

The bit of field-by-field walk through a certificate that nobody enjoys is that the answer changes depending on what you are trying to decide with it. Say what the decision is and the thread will converge.

Adding the measurement that post #10 says would settle it.

An itemised related-substances table with retention times says considerably more about the synthesis than a single total. A total tells you how much is not the main peak and nothing about what it is.

If it helps: the failure mode here is usually boring rather than dramatic.

Amino acid analysis gives content: it hydrolyses the peptide and quantifies the residues. It measures the peptide content independent of chromatographic purity. Very few supplier certificates report it.

I looked this up rather than remembered it, which is the right order.

Reading old certificates: if a certificate is dated years ago, the lot it describes might be old. That is not automatically a problem but it is worth noting. Fresh testing on the current lot is stronger than old testing on an older lot.

Where I part company with post #12, and it is a narrow parting.

Reframing field-by-field walk through a certificate slightly, because I think the disagreement is about the question rather than the answer. If the question is "does it happen", yes. If it is "how often", nobody here knows.

The arithmetic in post #14 is right; the assumption feeding it is the part to check.

Where the field-by-field walk through a certificate discussion usually stalls is that nobody wants to say "I do not know" and everyone is willing to say "it varies". Those are the same sentence with different clothes on.

A certificate of analysis is a statement by its issuer that a defined test was performed on a defined lot and produced a defined result. That is all it is, and it is not nothing. What makes it usable: lot identifier matching the container, test date, method identification specific enough to reproduce, the actual result as a number, and the acceptance criterion stated separately from the result.

That is what I would do. It may not be what is correct.

Endotoxin testing is separate: a certificate of purity says nothing about endotoxin. A chemically pure preparation can carry a clinically significant endotoxin load. If endotoxin matters, it needs to be tested and reported.

What fields matter on a certificate: lot number matching the vial, test date, the analytical method stated specifically, the measured result as a number, and the acceptance limit stated separately. A certificate missing any of these is weaker.

Worth separating two things that post #17 runs together.

Worth stating the null on field-by-field walk through a certificate before we explain it: the observation may be nothing. That possibility deserves a sentence and usually does not get one.

When method identifiers are missing: "reversed-phase HPLC" is less specific than "reversed-phase HPLC at 214 nm on a C18 column with a 10 to 40% acetonitrile gradient over 20 minutes". The second lets someone reproduce the analysis. The first does not.

Posting it because the silence on this was starting to look like agreement.

Post #25 is the version of this I will quote in future. One addition.

On field-by-field walk through a certificate I would separate what is worth knowing from what is worth acting on. The first list is long and the second is short, and conflating them is how threads get heated.

Answering the question post #25 raises rather than the one it answers.

"Not less than 98 per cent" is a specification. "98.3 per cent" is a result. A certificate carrying only the first has not told you what was measured.

Reading it again, the caveat matters more than the finding.

I had written a reply contradicting post #25 and deleted it. Here is what survived.

Peptide content and chromatographic purity are separate determinations and a certificate that reports only one has answered only one question. Content is the one that tells you how much is in the vial.

Not disagreeing with anyone above, just adding the bit I keep having to look up.

Offering a way to settle field-by-field walk through a certificate rather than another opinion about it. Two measurements, taken the same way, a fortnight apart. If the difference is within the noise, the question was not answerable at this precision.
