On the subject in the title: Storing lyophilised material long term: what governs the shelf life Working notes rather than a conclusion.

I have spent a fortnight trying to pin storing lyophilised material long term down and I want to set out where I have got to, because I suspect the honest answer is duller than the thread this will produce.

What I have: a consistent observation across a small number of cases, collected the same way each time. What I do not have: any controlled comparison, or any reason to think my cases are representative.

The specific question is whether the pattern survives once the obvious confounder is removed. I cannot remove it with what I have.

A methods point on storing lyophilised material long term rather than a substantive one: if the comparison is not like for like, the difference you are measuring is the difference in method.

The opening post and I disagree about the size of the effect, not about the direction.

If you are going to test stored material, test it against a sample of the same lot stored properly rather than against the certificate. Otherwise you are measuring the difference between two laboratories as well.

Practical answer on storing lyophilised material long term, since the theoretical one is upthread: do the simplest check first, write down the result, and only then decide whether the complicated explanation is needed. It usually is not.

Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed.

I have left out the parts I could not verify.

Protecting from light: amber vials or opaque containers reduce light exposure. If you are storing for long periods and light matters, this is worth doing. For short-term prepared solutions, ordinary storage away from direct sunlight is usually adequate.

Reading it again, the caveat matters more than the finding.

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

Adding the measurement that post #11 says would settle it.

Dry storage in a freezer is generally more protective than a refrigerator for long periods, provided the material is genuinely dry and the container genuinely sealed. Condensation on removal is the trap.

That matches what I was told, which is not the same as knowing it.

Door storage in a refrigerator is the warmest and most variable position in the appliance. A shelf at the back is a materially different environment.

The two questions that get merged here are whether material is degraded and whether it is contaminated. Storage affects both and they need different answers and different tests.

Picking up post #15: that is the part I would want checked first.

One of those cases where knowing the mechanism does not help the decision.

I read post #15 twice before replying, because I had assumed the opposite.

I would keep storing lyophilised material long term and the decision it usually gets used for separate in this thread. They are related and they are not the same question, and merging them is why the last one went badly.

Temperature excursions: if a vial sat at room temperature for a time, the question is whether the excursion was long enough to cause damage. Hours at room temperature is usually low risk. Days to weeks raises the risk. If you are unsure, contacting the supplier is more informative than guessing.

Storing lyophilised material long term: I would want to see the raw numbers rather than the summary before agreeing. Summaries lose exactly the information that would settle this.

Container choice matters: the material of the vial, the rubber closure, the fill volume all affect how quickly the contents degrade. Smaller fill volumes lose less to adsorption. Glass vials lose less to leaching than plastics.

I am reporting what happened, not recommending it.

Reframing storing lyophilised material long term slightly, because I think the disagreement is about the question rather than the answer. If the question is "does it happen", yes. If it is "how often", nobody here knows.

One more caveat and then I will stop qualifying: the sample selected itself.

I have three months of notes on storing lyophilised material long term and the honest summary is that the trend is real and the week-to-week numbers are noise. I nearly drew the opposite conclusion from the first fortnight.

Lyophilised material is far more stable than the same material in solution. That is why suppliers ship it dry, and it is the reason to reconstitute only what will be used.
