Gradient slope is the single biggest driver of apparent purity differences. A shallower gradient over a longer run resolves more impurities and gives a higher purity figure. A steep gradient produces a tidier-looking chromatogram with fewer visible peaks and gives a lower purity figure. Both are legitimate methods…

Where the integrator drew the baseline, and how much it moved the number — setting out what I have, and where I think it stops being reliable.

Posting the method first, because I know what the first three replies will otherwise be.

  • Column: C8, 4.6 x 150 mm, 3.5 um
  • Mobile phase: 0.1% TFA in water / 0.1% TFA in acetonitrile
  • Gradient: 13% to 57% organic over 22 minutes
  • Detection: 214 nm
  • Injection: 20 uL
  • Sample: tirzepatide, reconstituted to 0.5 mg/mL, injected within an hour

The main peak integrates at 98% of total area. There is a small feature on the trailing edge that I cannot decide is a shoulder or a baseline artefact, and that is what I am actually asking about.

This is why a purity figure without the underlying chromatogram is weaker evidence than it appears. It is also why two competent laboratories can report different numbers on the same vial without either being wrong.

Integrator would be much easier to settle if anyone reported the denominator. Almost nobody reports the denominator.

Coming back to post #3, because the follow-up matters more than the original answer.

I would rather this thread reach "we do not know" about integrator than reach a confident answer that nobody can support when asked.

Post #3 is right about the mechanism and I think understates the practical bit.

Area percent is a proportion of absorbance, not a proportion of mass. Two species with different extinction coefficients at the detection wavelength contribute unequally to the total, and nothing on the certificate corrects for that.

That is the version I use. It may not be the version that is correct.

Purity by chromatography answers "what proportion of what I detected is the intended species". It does not answer how much is in the vial, which is a separate assay.

Where the baseline is drawn is the least documented and most consequential choice in the whole determination. Valley-to-valley, tangent skim and forced-to-zero can span a couple of percentage points on the same chromatogram.

Worth reading the earlier posts in this thread before acting on mine.

Trifluoroacetic acid at 0.1 per cent is the near-universal ion-pairing additive for this work, and it also raises the baseline at 214 nanometres. That is why the same sample looks noisier at low wavelength.

That is one dataset and I would not build a rule on it.

Small correction to my own earlier position on integrator. I had the units the wrong way round, which changes the conclusion by an order of magnitude and therefore changes it entirely.

The question underneath integrator is usually "how would I tell?" rather than "what is true?", and that one has a method attached to it.

Write down what you would expect to see under each hypothesis before you collect anything. If they predict the same observation, collecting it will not help.

Answering the question post #10 raises rather than the one it answers.

A chromatogram image at a resolution where you can see peak shape but not baseline detail is worth having and is not the same as the data. Ask for the integration table if the number matters.

It took me longer than it should have to see that.

Column temperature affects retention and selectivity and is omitted from most certificates. Two runs at twenty-five and forty degrees are not the same method.

Adding the measurement that post #16 says would settle it.

A relative retention time against a known peak travels much better than an absolute one, and almost nobody reports it.

One of those cases where knowing the mechanism does not help the decision.

Before the thread moves on from integrator — what is the sample size behind the claim? I am not being difficult; I have seen the same figure quoted from an n of four and from an n of four hundred.

Worth separating two things that post #18 runs together.

On integration: where the baseline is drawn matters more than most people realise. On a clean chromatogram with well-resolved peaks the choice is inconsequential. On a chromatogram with a trailing shoulder or a rising baseline it matters. Differences of one to two percentage points between defensible integrations are ordinary.

If that is already documented somewhere, ignore me and link it.

I had written a reply contradicting post #20 and deleted it. Here is what survived.

Change the wavelength and the proportions change even though the sample has not. That is the reason the wavelength has to be on the document for the number to mean anything.

This has been discussed before and I could not find the thread, so, again.

Confirming post #20 from a second method, which matters more than confirming it from a second person.

An honest declaration on integrator: I have a prior here and it is strong enough that you should weight what I say downward. Stating it rather than hiding it.

Retention time is only comparable within a laboratory on a given method. Quoting a retention time across two reports as evidence of identity is not a valid comparison.

Where I would push back on the integrator consensus is the confidence, not the direction. The direction looks right. The confidence is borrowed.

Purity by chromatography answers "what proportion of what I detected is the intended species". It does not answer how much is in the vial, which is a separate assay.

It reads as pedantry until the day it does not.
