The arithmetic in the opening post is right; the assumption feeding it is the part to check.

Adding a reference point for titration planner refuses. Mine is a single case, collected without controls, and I am posting the method alongside it so it can be discounted appropriately.

Where I part company with the opening post, and it is a narrow parting.

Unit-conversion tools are only as good as the assumption they make about the syringe. Ask what barrel and what graduation interval it assumed before trusting the units figure.

Flagging that the sources on this are thinner than the confidence in the thread suggests.

A tool that refuses to give an answer when the result falls below one graduation is doing something useful. Most just print the number.

The number is defensible. The precision I gave it is not.

Narrowing post #3, because the general version has more than one answer.

The version of titration planner refuses that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Dose equivalence calculator: compounds have different potencies. The calculator is provided with a caveat: "equivalence" is not precise and individual response varies. It is a rough starting point, not a dose prescription.

I am confident about the direction and much less about the magnitude.

The most common input error is entering the vial mass in the concentration field. It produces a plausible number, which is why it survives.

Caveat: everything above assumes the paperwork is what it says it is.

Worth separating two things that post #10 runs together.

A titration planner is a scheduling aid rather than a clinical recommendation, and any tool that presents one as the other is overstepping.

The short version is the first sentence; the rest is why.

Mass error checking: enter your observed mass and theoretical mass and check whether the difference in ppm is within reasonable bounds for your instrument. Useful sanity check on LC-MS results.

Reading it back, the second half matters more than the first.

Post #16 is right about the mechanism and I think understates the practical bit.

What I would tell a new member reading about titration planner refuses for the first time: the confident posts are not the reliable ones, and the reliable ones are longer.

Coming back to post #15, because the follow-up matters more than the original answer.

A saved calculation is worth more than a remembered one. Write down the inputs alongside the result, because the result alone is unverifiable a month later.

A partial answer, offered because a partial answer beats none.

Taking post #16 at face value and following it one step further.

Small methodological point on titration planner refuses: repeating a measurement is cheap and resolves most of what is being argued about here at no cost to anyone.

Where I would push back on the titration planner refuses consensus is the confidence, not the direction. The direction looks right. The confidence is borrowed.

Significant figures in the output should not exceed what the inputs support. A concentration calculated from a nominal vial mass is not known to four decimal places.

The confident version of this sentence would be wrong, so here is the hedged one.

Precision and significant figures: the calculators do not round to match your input precision. If you input values to one decimal place, the output might have more. Use significant figures appropriate to your input precision.

Worth reading the earlier posts in this thread before acting on mine.

Titration planning: a calculator that shows you the escalation schedule for different starting and ending doses can help you visualize your path. Actual path might be slower or adjusted for tolerability.

I would put the burden of proof on the interesting explanation, not the dull one.

Worth separating titration planner refuses as a question about the compound from titration planner refuses as a question about the documentation. They get answered by different people and only one of them is answerable here.

Confirming post #27 from a second method, which matters more than confirming it from a second person.

Where I have landed on titration planner refuses, having got it wrong once in public: the direction is clear, the magnitude is not, and anyone quoting a precise magnitude has borrowed it from somewhere that did not measure it.

Answering the question post #27 raises rather than the one it answers.

Rounding behaviour is where two tools disagree. One rounding to the nearest graduation and one to two decimal places will produce different advice from identical inputs.
