Posting this under the heading it deserves: Second pass at: Cost versus information: making the trade-off explicitly Everything below is what sits behind that.

Posting a small dataset on Cost versus information. It is mine, it is uncontrolled, and the method is stated so it can be discounted appropriately.

What I would like is not agreement but a second dataset collected by someone with no stake in mine. If one exists I would rather read it than argue for this one.

Reconstitution behaviour is a legitimate home observation. A cake that will not dissolve, a solution that hazes, visible particulate — all are real information and all are free.

Where the Cost versus information reasoning breaks down for me is the step from the group result to the individual case. That step is almost never argued for.

A note on scope: what I am saying about Cost versus information applies to the case in the first post and I would not extend it further without checking.

A home refractometer is not measuring what people hope it is measuring in these preparations, and the readings are dominated by everything other than the peptide.

I think the Cost versus information question is answerable and has not been answered, which is a more optimistic position than most of this thread.

Post #6 describes the usual case. This is about the unusual one.

Cost versus information would be much easier to settle if anyone reported the denominator. Almost nobody reports the denominator.

Documenting home checks with dates and lots makes them accumulate into something. Undocumented ones evaporate and get remembered selectively.

The honest cost comparison is a home kit against a single independent submission. For most people the second answers the question and the first answers a different, smaller one.

It is worth checking rather than assuming, which costs nothing.

What I would tell a new member reading about Cost versus information for the first time: the confident posts are not the reliable ones, and the reliable ones are longer.

Cost versus information has been discussed here with more heat than it deserves, mostly because two definitions have been in play the whole time.

The right question for any home test is: what physical quantity does it measure, and what would have to be true for that quantity to answer your question? Most disappointment comes from skipping it.

It is worth stating the boring hypothesis before the interesting one.

Cost versus information sits at the boundary between what this community can usefully discuss and what it cannot, and I think it falls on the discussable side, narrowly.

Answering the question post #13 raises rather than the one it answers.

Ultraviolet absorbance at 280 nanometres estimates concentration for peptides containing aromatic residues and gives nothing for peptides that do not. Knowing which yours is comes first.

It is a small point and it changes the answer, which is an awkward combination.

Post #17 and I disagree about the size of the effect, not about the direction.

An honest declaration on Cost versus information: I have a prior here and it is strong enough that you should weight what I say downward. Stating it rather than hiding it.

Cost versus information is well covered in the tag pages, and the older discussions are better than the recent ones because they were argued out properly. Worth twenty minutes before adding to this one.

Where I part company with post #20, and it is a narrow parting.

The reason Cost versus information keeps being re-asked is that the answer is conditional and people quote it without the condition. It is not that the answer is unknown.

What you can use home tests for: confirmation that a solution contains peptide (as opposed to being saline alone), rough screening for very different compounds (semaglutide versus totally unrelated compound). What you cannot use them for: precise purity assessment or verification that the identity is correct.

Nobody has said the unglamorous part of Cost versus information yet, so: most of the variation is explained by things that are boring to write about and easy to check.

My experience of Cost versus information contradicts the reply above. I am posting it as a data point rather than as a refutation, because one person's experience is exactly that.
