Narrowing the opening post, because the general version has more than one answer.

Detection wavelength: 214 nm detects the peptide bond and is relatively insensitive to composition. 280 nm detects aromatic residues and is strongly composition-dependent. Area percent at one wavelength is not area percent at the other.

I would treat the number as indicative rather than as a measurement.

What I would check first on system suitability injections is whether the thing being measured moved or whether the way of measuring it moved. Those look identical in a graph.

Taking post #4 at face value and following it one step further.

Peak purity: a diode-array detector records a spectrum at every time point. If a peak contains two co-eluting species with different spectra, the spectrum changes across the peak. A passing peak-purity result says the spectrum is constant; it is weak evidence of homogeneity if the impurities have similar spectra.

That is where I would start, not where I would stop.

I would be cautious about generalising from the system suitability injections example above. It is a good example. It is one example.

Gradient slope is the single biggest driver of apparent purity differences. A shallower gradient over a longer run resolves more impurities and gives a higher purity figure. A steep gradient produces a tidier-looking chromatogram with fewer visible peaks and gives a lower purity figure. Both are legitimate methods and they will not produce the same number.

What is documented is narrow. What is inferred from it is broad. The gap between them is where every argument here lives.

This follows post #8 rather than contradicting it.

System suitability testing: injections run before and during the sample run to establish whether the instrument, column and method were performing when the sample was analysed. If suitability did not pass, the sample results from that run are uninterpretable.

System suitability injections: I would want to see the raw numbers rather than the summary before agreeing. Summaries lose exactly the information that would settle this.

Practical note on system suitability injections: write down what you expect before you look. The number of times I have found what I went looking for is higher than chance would allow.

Where the baseline is drawn is the least documented and most consequential choice in the whole determination. Valley-to-valley, tangent skim and forced-to-zero can span a couple of percentage points on the same chromatogram.

System suitability injections is one of those subjects where the general answer and the answer for a specific case diverge, and the thread will go in circles until someone says which one is being asked for.

If two laboratories disagree by more than two or three percentage points, work through method, integration, sample handling, whether it was the same lot and the same vial, and whether suitability passed. After all five, a gap needs an explanation.

System suitability is not paperwork. If the replicate injections failed, the run did not happen — the numbers from it are uninterpretable rather than approximate.

That is the version I would defend. It is not the version I started with.

Reading the surrounding paragraph is worth the two minutes. The authors are more careful than their summarisers.

A chromatogram image at a resolution where you can see peak shape but not baseline detail is worth having and is not the same as the data. Ask for the integration table if the number matters.

A partial answer, offered because a partial answer beats none.

Post #17 is right about the mechanism and I think understates the practical bit.

On system suitability injections, the part that usually goes wrong is that the question is asked as though it has one answer. It has a range, and the width of the range is the interesting bit.

If you can post the two or three numbers you are working from, several people here will check the arithmetic rather than argue about the conclusion.

Typical suitability criteria are a replicate area relative standard deviation below about two per cent, a tailing factor inside a defined window, a resolution minimum against a specified peak, and a plate-count floor.

The short version is the first sentence; the rest is why.

Particle size and column dimensions determine what resolution is achievable at all. A 5 micrometre 250 millimetre column and a sub-2 micrometre 100 millimetre column are different instruments in practice.

I would want to see it done twice before believing it once.

Peaks that do not elute do not appear in the area percentage. Aggregates and strongly retained species can be entirely invisible to a standard method, which is a ceiling on what any purity figure can claim.

I would put this at better than even and not much better.

Gradient delay volume differs between instruments and shifts the whole chromatogram. It is why a transferred method rarely reproduces retention times exactly on a different system.

Worth separating two things that post #24 runs together.

One more thing on system suitability injections that took me far too long to see: the two figures people quote are not measuring the same quantity. Once you notice that, the apparent contradiction disappears.
