I had written a reply contradicting post #89 and deleted it. Here is what survived.

Transfer between laboratories: a method can be transferred from one lab to another, but the receiving lab needs to demonstrate that they can achieve the same performance. This requires comparative testing and sometimes small method refinements.

If it helps: the failure mode here is usually boring rather than dramatic.

Forced degradation studies: deliberately stress the material with acid, base, oxidant, heat, light to generate degradation products and demonstrate that the method can separate them from the parent peak. Acceptance is that the method is stability-indicating.

Picking up post #93: that is the part I would want checked first.

Taking forced degradation studies seriously for a moment rather than deflecting: the honest position is that the community has observations and no controlled comparison, and those two things support very different sentences.

Forced degradation under acid, base, oxidation, heat and light is how specificity is actually demonstrated. The chromatograms from that work are more informative than the release data.

That holds under the stated conditions and I have stated them.

Nobody has said the unglamorous part of forced degradation studies yet, so: most of the variation is explained by things that are boring to write about and easy to check.

Post #93 describes the usual case. This is about the unusual one.

Forced degradation studies is well covered in the tag pages, and the older discussions are better than the recent ones because they were argued out properly. Worth twenty minutes before adding to this one.

Adding the measurement that post #97 says would settle it.

The most useful single question about a method: what would it fail to detect? Every method has an answer and few documents state it.

I looked this up rather than remembered it, which is the right order.

The version of forced degradation studies that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Post #97 put the caveat in the right place and I want to underline it.

System suitability is the ongoing evidence that a validated method is still performing. Validation is done once; suitability is done every run, and it is the one that appears on a certificate.

Adding it in case it saves somebody the afternoon it cost me.

Why two laboratories may disagree: after validating the same method, they may still report different purity on the same sample due to integration differences, column age differences, subtle differences in mobile phase pH or temperature. On forced degradation studies: the maintained page in the documentation commons covers the general case with citations and a review date, which is more reliable than any reply here including this one.

Range and working range are different things and a certificate rarely distinguishes them. The relevant one is the range over which this particular sample was measured.

If the premise is wrong, everything after it is decoration.

Taking post #105 at face value and following it one step further.

Where I have landed on forced degradation studies, having got it wrong once in public: the direction is clear, the magnitude is not, and anyone quoting a precise magnitude has borrowed it from somewhere that did not measure it.
