Adding a reference point for Lyophilisation. Mine is a single case, collected without controls, and I am posting the method alongside it so it can be discounted appropriately.

Lyophilisation and why the cake looks the way it does Writing it up because I had to work it out twice and would rather nobody else did.

I was wrong about Lyophilisation in a thread last spring and I would like to correct it publicly rather than quietly.

The error was in the units, which changed the conclusion by an order of magnitude. Setting out the corrected version, and the way I now check for that class of mistake.

I had written a reply contradicting the opening post and deleted it. Here is what survived.

The version of Lyophilisation that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Confirming the opening post from a second method, which matters more than confirming it from a second person.

A collapsed cake indicates the product warmed above its collapse temperature during drying. It is not necessarily a quality failure and it is a real observation about the process.

Caveat: everything above assumes the paperwork is what it says it is.

The opening post describes the usual case. This is about the unusual one.

Lyophilisation and reconstitution: the powder is stable for extended periods if kept cool and dry. Once reconstituted, stability decreases because the dissolved peptide is more vulnerable to oxidation and other degradation.

I would rather say I do not know than round it up to an answer.

Post #5 is the version of this I will quote in future. One addition.

Antioxidants: compounds that prevent oxidation (which damages peptides) are sometimes included. Methionine oxidation is a common degradation pathway and antioxidants help prevent it.

That is all I can say without guessing.

The arithmetic in post #8 is right; the assumption feeding it is the part to check.

Bulking agents such as mannitol give the cake structure and contribute to the mass in the vial. Their presence is one reason declared fill and peptide content are different numbers.

It is the kind of thing that is obvious once and never again.

What I can speak to on Lyophilisation is narrow, so I will keep it narrow rather than generalising from it. Beyond that boundary I do not know.

Coming back to post #8, because the follow-up matters more than the original answer.

Since Lyophilisation keeps coming up, it should probably be a maintained page rather than a recurring thread. I am happy to draft it if someone with more direct experience will review it.

Benzyl alcohol as a preservative is also a solubiliser for some peptides, which is an incidental property that occasionally matters.

Buffer choice affects both solubility and stability, and the two do not always point the same way. A formulation is a compromise between them rather than an optimum for either.

Post #14 answers the question as asked. The question underneath it is different.

On Lyophilisation I would separate what is worth knowing from what is worth acting on. The first list is long and the second is short, and conflating them is how threads get heated.
